Research achievements
As of April 28, 2026, Dr. Casola’s research activity is documented by 69 articles and reviews published in peer-reviewed international journals. His research focuses on the genetic and epigenetic mechanisms that regulate B-cell development and immune function in both physiological and pathological conditions. In parallel, Dr. Casola leads a research program aimed at identifying the molecular determinants underlying B-cell lymphoma initiation, progression, and therapy resistance.
These studies are conducted using genetically engineered mouse models (many developed within his laboratory) as well as primary mouse and human normal and malignant B cells. Clinical samples are obtained through collaborations with internationally recognized pathologists and hemato-oncologists.
Dr. Casola has made the following major contributions:
• Generated the first transgenic mouse model enabling conditional gene targeting in B cells participating in the germinal center reaction during adaptive immune responses (Casola et al., PNAS, 2006; corresponding author).
• Developed mouse models to investigate the role of Epstein-Barr Virus proteins LMP1 and LMP2A in the pathogenesis of Hodgkin disease and post-transplant EBV-associated lymphoproliferative disorders (Casola et al., Nature Immunology, 2004, corresponding author; Zhang, Casola et al., Cell, 2012, first author).
• Identified microRNA miR-155 as a key regulator of germinal center B-cell responses (Thai et al., Science, 2006; co-author).
• Demonstrated the role of the Polycomb group protein Ezh2 in transcription factor–mediated somatic cell reprogramming (Fragola et al., 2009; corresponding author).
• Elucidated the function of the Polycomb group protein Ezh2 in B-cell adaptive immune responses (Caganova et al., 2013; corresponding author).
• Applied somatic nuclear transfer technology to demonstrate the nuclear plasticity of terminally differentiated antibody-secreting plasma cells (Kumar et al., PNAS, 2015; corresponding author).
• Defined the contribution of VH gene replacement to diversification of the primary mouse immunoglobulin heavy chain repertoire (Kumar et al., PNAS, 2015; corresponding author).
• Revealed the existence of mouse and human Burkitt lymphomas bypassing B-cell receptor (BCR) requirement, providing mechanistic insights into BCR function in MYC-driven lymphomas (Varano et al., Nature, 2017; corresponding author).
• Reconstructed the molecular mechanisms underlying the origin of high-grade B-cell lymphomas with MYC and BCL2 rearrangements (Varano et al., Blood Cancer Discovery, 2025; corresponding author).
• Establishewd the role fo the RAG proteins in the pathogenesis of high-grade B-cell lymphomas with MYC and BCL2 rearrangements (Varano et al., Blood Cancer Discovery, 2025; corresponding author).
• Identified the putative precursor cell of origin for high-grade B-cell lymphomas with MYC and BCL2 rearrangements (Lonardi et al., Leukemia, 2026; corresponding author).

Since his appointment in 2006 as Principal Investigator at IFOM, Dr. Casola has secured continuous competitive funding from major national and international agencies, including the European Research Council (ERC), Horizon 2020, Worldwide Cancer Research (formerly AICR), the Italian Association for Cancer Research (AIRC), the Telethon Foundation, the CARIPLO Foundation, and the Italian Ministry of Health.
Dr. Casola was also awarded a collaborative research grant with Dr. S. Fagarasan (RIKEN Center for Integrative Medical Sciences, Yokohama, Japan), supported by RIKEN.

Over the past 20 years, Dr. Casola’s research unit has established collaborations with leading international groups in B cell biology, lymphoma research, and epigenetics. These include laboratories led by K. Rajewsky (Max Delbrück Center, Berlin), R. Dalla-Favera and L. Pasqualucci (Columbia University, New York), R. Siebert (Ulm University), U. Klein (University of Leeds), M. Schmidt-Supprian (Technical University of Munich), M. Busslinger (Institute of Molecular Pathology, Vienna), D. Hodson (University of Cambridge), T. Kurosaki (Osaka University), S. Fagarasan (RIKEN, Yokohama), D. Kitamura (Tokyo University of Science), M. Hikida (Akita University), and M. Maruyama (National Center for Geriatrics and Gerontology, Nagoya).
