Abstract:
Bovine adrenal gland contains a large amount of ME-like peptides which are preferentially localized in medulla. Here, three different molecular forms of the ME-like peptides have been detected by gel filtration chromatography developed with 1 N CH3COOH on 6 M guanidine and by antiserum-sepharose immunoabsorption experiments. A low molecular weight form is identified as ME by its chromatographic characteristics and susceptibility to CNBr. The high molecular weight forms of ME-like peptides have little or no affinity for opiate receptors, but when digested with trypsin they generate smaller peptides with high affinity for opiate receptors. We could not detect BLPH (61-69) which is obtained following tryptic digestion of BLPH (61-91) thus excluding that these high molecular weight ME-like peptides are α, β, or γ-endorphin. One of the products of the tryptic cleavage was distinguishable from ME or BLPH (61-65) and another tryptic peptide was identical to ME in the gel filtration, thin layer chromatography and susceptibility to proteolysis. In conclusion, some of the multiple forms of ME-like peptides found in adrenal medulla have high molecular weight, contain the ME sequence within their structure and can form ME following tryptic digestion; thus, they have many of the properties expected for ME precursors.
